(Eisai Sensei)
(Eisai Sensei)
(DailyMed)
(DailyMed)
(DailyMed)
(Eisai Sensei)
(Eisai Sensei)
(DailyMed)
(DailyMed)
(DailyMed)
(DailyMed)
(DailyMed)
Silodosin
(DailyMed)
(Eisai Sensei)
(Eisai Sensei)
Silodosin 4 mg per film-coated tablet
(Eisai Sensei)
8 mg/day. (Eisai Sensei)
(Eisai Sensei)
(Eisai Sensei)
(Eisai Sensei)
(Eisai Sensei)
(DailyMed)

DailyMed (DailyMed)
Silodosin 4 mg is a highly selective α1A-adrenoceptor antagonist used to relieve lower urinary tract symptoms associated with benign prostatic hyperplasia (BPH). It relaxes smooth muscle in the prostate, bladder neck, and prostatic urethra, thereby improving urinary flow. (Eisai Sensei)
Silodosin selectively antagonises α1A-adrenergic receptors, which are highly expressed in the prostate, bladder neck, and prostatic urethra.
Blockade of these receptors relaxes smooth muscle and reduces bladder outlet resistance, thereby improving both voiding and storage symptoms associated with BPH. (European Medicines Agency (EMA))
None.
Silodosin has no antibacterial, antiviral, or antifungal activity.
Silodosin is absorbed following oral administration. In internationally studied formulations, absolute bioavailability is approximately 32%, with peak plasma concentrations generally reached within approximately 2–3 hours.
Silodosin is highly protein bound and is metabolised through several pathways, including UGT2B7, alcohol and aldehyde dehydrogenases, and CYP3A4.
The terminal elimination half-life is approximately 13 hours in international prescribing information. (DailyMed)
Silodosin is contraindicated in patients with known hypersensitivity to Silodosin or any component of the formulation.
International product information also contraindicates or does not recommend use in patients with severe renal impairment or severe hepatic impairment, depending on the regulatory jurisdiction. (DailyMed)
Concomitant use with strong CYP3A4 inhibitors should generally be avoided where specified in the applicable product labeling. (DailyMed)
Silodosin may cause postural or orthostatic hypotension, dizziness, and occasionally syncope, particularly after initiation of therapy. (Eisai Sensei)
A characteristic adverse effect is abnormal ejaculation, including markedly reduced ejaculate volume or absence of semen during orgasm. This effect is generally reversible after discontinuation.
Patients undergoing cataract surgery should inform the ophthalmic surgeon of current or previous Silodosin use because Intraoperative Floppy Iris Syndrome (IFIS) has been reported with α1-adrenergic antagonists. (DailyMed)
Silodosin is indicated for men with BPH and is not indicated for use in women.
Pregnancy and lactation therefore do not form part of its approved standard clinical use. (European Medicines Agency (EMA))
The most characteristic adverse reaction is ejaculatory dysfunction, including reduced or absent ejaculation.
Other adverse effects may include dizziness, orthostatic hypotension, headache, nasal congestion, diarrhoea, and nausea.
Rare clinically important reactions include syncope, hypersensitivity reactions, and angioedema. (DailyMed)
The principal expected effect of overdose is hypotension resulting from α1-adrenoceptor blockade.
Management is supportive, including supine positioning, cardiovascular support, intravenous fluids, and vasopressors when clinically indicated.
Because Silodosin is highly protein bound, dialysis is unlikely to be effective in substantially enhancing elimination. (DailyMed)
Strong CYP3A4 inhibitors, including ketoconazole, itraconazole, clarithromycin, and ritonavir, may substantially increase Silodosin exposure. (DailyMed)
Strong P-glycoprotein inhibitors may also increase exposure.
Concomitant use with other alpha-blockers is generally not recommended because of additive hypotensive effects.
Use with PDE5 inhibitors such as sildenafil or tadalafil may increase the risk of dizziness or hypotension and requires caution. (Eisai Sensei)
Antihypertensive medicines may also produce additive blood-pressure lowering.
Silodosin 4 mg per film-coated tablet
The specified EISAI-Urief 4 mg 3×10 tablets corresponds to URIEF (TABLETS 4 MG) registered in Thailand as a Silodosin 4 mg tablet. Thai market information lists a 3 × 10 tablet pack.
Indicated for bladder outlet obstruction and lower urinary tract symptoms associated with benign prostatic hyperplasia (BPH). (Eisai Sensei)
Silodosin does not directly reduce prostate size in the way that Finasteride or Dutasteride does; rather, it improves symptoms primarily by reducing smooth-muscle tone and bladder outlet resistance.
For URIEF 4 mg marketed in Thailand, Eisai product information specifies:
Silodosin 4 mg orally twice daily after breakfast and the evening meal, giving a total dose of 8 mg/day. (Eisai Sensei)
The dosage may be reduced according to the patient’s clinical condition, particularly when tolerability, renal function, hepatic function, or blood pressure is a concern. (Eisai Sensei)
This is an important product-specific point: the Thai/Japanese URIEF regimen is 4 mg twice daily, whereas several European and U.S. Silodosin products use 8 mg once daily with food. The Thai URIEF prescribing information should therefore be followed for this specific product. (Eisai Sensei)
Silodosin may cause orthostatic hypotension, dizziness, or syncope, particularly in patients who are older, have low baseline blood pressure, or are receiving antihypertensive or PDE5-inhibitor therapy. (Eisai Sensei)
Patients should be informed about the relatively common occurrence of ejaculatory dysfunction, including reduced or absent ejaculation.
Patients scheduled for cataract surgery should inform their ophthalmologist of current or previous Silodosin use because of the risk of Intraoperative Floppy Iris Syndrome (IFIS). (DailyMed)
Store in the original packaging, protected from moisture and light. Follow the specific storage temperature stated on the Thai URIEF 4 mg product labeling.
