(Medicines.org.uk)
PBC. (Medicines.org.uk)
Ursodeoxycholic Acid
(Medicines.org.uk)
(DailyMed)
(Medicines.org.uk)
(DailyMed)
(Medicines.org.uk)
(Medicines.org.uk)
(Medicines.org.uk)
(Medicines.org.uk)
UDCA. (Medicines.org.uk)

Ursodeoxycholic Acid 250 mg is a naturally occurring hydrophilic bile acid used mainly in the treatment of primary biliary cholangitis (PBC) and for the dissolution of suitable cholesterol gallstones in patients with a functioning gallbladder. (Medicines.org.uk)
Ursodeoxycholic Acid, or UDCA, alters the composition of the bile acid pool toward less hydrophobic and less hepatotoxic bile acids.
It also reduces intestinal cholesterol absorption and cholesterol secretion into bile, thereby decreasing biliary cholesterol saturation and facilitating the gradual dissolution of susceptible cholesterol gallstones.
In cholestatic liver disease, UDCA also exerts cytoprotective effects on hepatocytes and cholangiocytes. (Medicines.org.uk)
None.
Ursodeoxycholic Acid is not an antibiotic, antifungal, or antiviral agent and has no direct antimicrobial activity.
Ursodeoxycholic Acid is absorbed from the gastrointestinal tract and undergoes extensive enterohepatic circulation.
After hepatic uptake, it is mainly conjugated with glycine and taurine and secreted into bile. During continued therapy, UDCA becomes a progressively larger component of the circulating bile acid pool. (Medicines.org.uk)
Ursodeoxycholic Acid is contraindicated in patients with known hypersensitivity to the drug or any component of the formulation.
U.S. prescribing information also contraindicates its use in patients with complete biliary obstruction. (DailyMed)
For gallstone dissolution, treatment is generally inappropriate in patients with calcified or radiopaque gallstones, impaired gallbladder contractility, or frequent biliary colic. (Medicines.org.uk)
Liver function should be monitored during treatment, particularly in patients with PBC.
The U.S. label recommends monitoring γ-GT, alkaline phosphatase, AST, ALT, and bilirubin monthly for the first 3 months and every 6 months thereafter. (DailyMed)
Patients treated for gallstone dissolution should undergo appropriate imaging follow-up to assess treatment response and detect gallstone calcification. (Medicines.org.uk)
Use during pregnancy should be based on the indication and an individualized assessment of benefit and risk.
Women receiving UDCA specifically for gallstone dissolution should use effective contraception; some SmPCs recommend a non-hormonal contraceptive method because hormonal contraception may promote gallstone formation. (Medicines.org.uk)
Breastfeeding decisions should be individualized according to the clinical indication and medical advice.
The most commonly reported adverse effect is loose stools or diarrhea.
In patients with PBC, pruritus may temporarily worsen at the beginning of treatment. In such cases, dose reduction followed by gradual re-escalation may be considered. (Medicines.org.uk)
Rare cases of hepatic decompensation have been reported in patients with advanced cirrhosis. (Medicines.org.uk)
The most likely manifestation of overdose is diarrhea.
Management is supportive, with correction of fluid and electrolyte losses if necessary.
Bile acid-binding agents such as cholestyramine and colestipol, as well as certain aluminium-containing antacids, may reduce the absorption of Ursodeoxycholic Acid.
Medicines that increase biliary cholesterol secretion, including some estrogen-containing therapies, may reduce the effectiveness of UDCA for gallstone dissolution. (Medicines.org.uk)
Ursodeoxycholic Acid 250 mg
The product specified is an oral 250 mg hard capsule, supplied in a 10 × 10 capsule pack. Thai regulatory data confirm this strength and dosage form.
Ursodeoxycholic Acid is indicated for the treatment of Primary Biliary Cholangitis (PBC).
It is also used for the dissolution of suitable radiolucent cholesterol gallstones in patients with a functioning gallbladder. (Medicines.org.uk)
Thai product information additionally lists prevention of gallstone formation in obese patients undergoing rapid weight loss. (Made in Thailand)
For PBC in adults, the U.S. prescribing information recommends 13–15 mg/kg/day, administered in 2–4 divided doses with food. (DailyMed)
European SmPCs commonly use approximately 12–16 mg/kg/day, depending on body weight and disease stage. (Medicines.org.uk)
For cholesterol gallstone dissolution, approximately 8–12 mg/kg/day is commonly recommended, with some SmPCs advising administration in the evening or at bedtime. (Medicines.org.uk)
The 250 mg capsule strength does not mean that the usual therapeutic dosage is 250 mg once daily for every indication.
UDCA does not dissolve all types of gallstones and should not be used in patients with complete biliary obstruction, calcified gallstones, or markedly impaired gallbladder function. (DailyMed)
Patients with PBC require periodic liver-function monitoring. Clinical deterioration, worsening jaundice, ascites, confusion, or other signs of hepatic decompensation require prompt medical assessment.
Store in the original package in a dry place, protected from excessive heat and moisture. The storage temperature stated on the labeling of the specific Ursodeoxycholic Acid 250 mg capsule product should be followed.
